Observation of epithelial tissue

Objective: Learn to prepare a sample for the epithelium from the inside the mouth.
                   Observe the sample with the microscope.

MATERIALS

-          Microscope.
-          Slide and cover slip.
-          Staining tray.
-          Staining support.
-          Washing bottle. 

Chemical coolorant: methylene blue.

ACTIVITIES

     1. Observe the sample with the lowest magnification lens and draw what you see.
     2. Identify the main cell organelles in your drawing and write their names.
3. Change the lens to one or greater magnification and observe the preparation again. Explain what differences you can see.

The muscular system. The muscles. Striated muscle tissue

OBJECTIVES
1. Observe chicken’s muscle cells under the optical microscope;
2. Draw the muscular cells.

MATERIALS

- Microscopes.
- Needles.
- Slides and cover slips.
- Staining trays.
- Staining support.
- Bottle washer.

Chemicals and reagents: eosin and 50% glycerine (optional).


PROCEDURE

1. Take a small piece of muscle about 5 mm wide and 2 mm thick. Place it on the slide with a few drops of saline solution.
2. Try to cut the muscle with the needles. One edge has to become similar to a brush. To prevent the muscle from drying, add the saline solution if necessary.
3. After achieving disintegration, put the covers on the slide and observe it with the microscope. Look for the area of ​​greatest disintegration and observe it with the lens of grater magnification.


QUESTIONS:

1. What is the appearance of cells? Are they round or elongated?


2. Where is the nucleus located?
 

3. What is the function of this tissue? 

4. Do they contrac in a voluntary or involuntary way?

5. What is the function of skeletal muscles?



6. Draw the cells you have observed with a total magnification of 100X or 400X. Focus on the internal cell structure and write it down on your picture.
